Cryo-EM sample preparation tool with extended run time and enhanced automation
Aquilos 2 Cryo-FIB is the latest generation of Thermo Scientific cryo-dual beam systems. It is dedicated to the preparation of thin, electron-transparent lamellas for high-resolution cryo-electron tomography or MicroED of micro-crystals. The Aquilos 2 Cryo-FIB allows users to take a revolutionary step forward by automating the production of multiple lamellas from specific programmed locations. Longer run times and low contamination rates will be the advantages provided by the dedicated cryo-hardware.

- You can allow the system to automatically produce multiple lamellas with Thermo Scientific AutoTEM Software by running it overnight
- Using Thermo Scientific Auto Slice and View Software, discover native subcellular structures below the surface and identify features for high-resolution tomography in cryo-TEM
- Even for overnight runs, your samples remain at vitreous temperatures. The use of cryohardware and a fully rotatable cryostage ensures that frozen samples remain vitrified throughout your experiments
- Multiple cells can be marked for cryo-lamella milling. Cryo AutoTEM Software prepares lamellas in an unattended manner using guided process instructions, increasing productivity
- Prepare lamellas with nanometer-position accuracy from specific targeted regions
- You can obtain 3D images from vitrified samples by sequentially milling and then imaging the cross-section of a defined volume of the sample using the Cryo-Auto Slice and View Software. Using this procedure, you can stop milling at the correct subcellular region to prepare your lamella before you observe the ultrastructural detail using cryo-TEM
- Using the iFLM Correlative System, samples can be imaged directly within the high vacuum of the Aquilos 2 Cryo-FIB, without having to transfer them from an external cryo-light microscope. The process saves time and reduces the risk of contamination. Fluorescently marked cells can be found directly in the Aquilos
- Thermo Fisher cryo-electron tomography offers a workflow that covers flash-freezing cells to 3D visualization. Interior cellular regions are imaged at nanometer-scale resolution and from cryo-lamellae precisely prepared with Aquilos 2 cryo-FIB
